Since yesterady / 13th of December) I am not able to submit any ticket nor can I save any comments or answer a customer.
The "button" is not there anymore. We made no changes and I am the only one having this problem within our organisation. The error occured by accident in the afternoon.
Changed the browser (updated to newest versions), cleared cache, logged out an in again

When you say the Button is not there, are you referring to the bottom right Status where you can submit the ticket as Open, Pending, Solve, etc? Is it not present at all or just greyed out? Are you the assignee of the ticket when attempting to make the change?
I would try a few basic troubleshooting steps to see if it helps such as:
1. Do a full clearing of your Browser Cache.
2. Try from a secondary browser or an Incognito window if using Chrome.
3. Try disabling any background apps or extensions from your Browser. When you are within the ticket try using this URL https://YOURSUBDOMAIN.zendesk.com/agent/tickets/12345?noapps and replace the "YOURSUBDOMAIN" with your actual ZendesK SUBDOMAIN as well as replacing the ticket number with the ticket you are on and then refresh the browser.
