I am wanting to remove the predefined "-" that is the default value in the dropdown field. I would like to replace it with "not specified"

As far as I'm aware there is no way to remove the dash which you're looking to remove here. The purpose of the dash is to indicate to the user filling in the field, that the current field is empty and requires a selection.
I realise this may not be an ideal answer to your question here but I hope it helps in some way. :)

We would also see value in being able to remove this - option from a custom drop down menu. We are wanting our customers to choose from 5 possible options from the custom drop down menu and we dont want to make that choice for them/default to one of these 5 options, so we have no other choice other than to choose setting the default selection to - . Is it not possible to make a drop down a required field and the only options the customer can choose are the ones listed against the drop down menu?

You can make a drop-down a required field for your customers. You just need to select the "Require to submit a request" option in the drop-down field settings. More information here
However, the "-" option can't be removed as this is a default value that is hard-coded in the system.
