Okay, I have a question does the report measures the data after business hours as well? Like if an update is given by a customer after business hours?

That really depends on what report or query you're looking at. Some will not segment data by business hours, others will, and some give you the option for both. Here's a few examples:

So basically we wanna calculate the amount of customer updates on specific tags, Now my question is will the report update if we a customer update is after business hours?

Yes we use explore for specific tags for getting customer updates as that how many customer have given response to our sample emails.
