Hi,
i was wondering if it was possible to add a custom user field like "Title" and have it added to the agents signatures in Settings > Agents > Signature
Or do we have to use Personal Agent Signature to add that kind of custom information?.

You'd most likely need to use the personal agent signature field to have titles show up next to the agents name.
Otherwise, you could maybe create a custom user field with the job title.
Then add a signature to your Notify Requester triggers using the available placeholders
